The magnetic proton recoil(MPR)spectrometer is a novel diagnostic instrument with high perfor-mance for measurements of neutron spectra in inertial confinement fusion(ICF)experiments and high power fusion devices.A compact MPR-type spectrometer dedicated to the research of pulsed deuterium-tritium(DT)neutron spectroscopy of special experimental conditions is currently under design.Analyses of the main parameters and performance of the magnetic analysis system through 3-D particle transport calculations and MonteCarlo simulations and calibration of the system performance as a test using CR-39 solid track detector and α particle from 239pu and 226Ra radioactive sources are presented in this paper.The results indicate that the magnetic analysis system will achieve a detection efficiency level of 10-5-10-4 at an energy resolution of 1.5%-2.1%,and fulfills the design goals of the spectrometer. 相似文献

An artificial site‐selective DNA cutter to hydrolyze single‐stranded DNA at a desired site was prepared from CeIV/ethylenediamintetraacetic acid (EDTA) and two ethylenediamine‐N,N,N′,N′‐tetrakis(methylenephosphonic acid)–oligonucleotide conjugates. By using this cutter, the sense strand of a blue fluorescent protein (BFP) gene was selectively cut at a predetermined site in the chromophore‐coding region. The upstream fragment obtained by the site‐selective scission was ligated with the downstream fragment of the closely related green fluorescent protein (GFP) gene so that the 5′‐ and 3′‐end portions of the chromophore came from the BFP fragment and the GFP fragment, respectively. The recombinant gene was successfully expressed in E. coli and the chimeric chromophore emitted green fluorescence as expected. 相似文献

Biomolecular droplets formed through phase separation have a tendency to fuse. The speed with which fusion occurs is a direct indicator of condensate liquidity, which is key to both cellular functions and diseases. Using a dual-trap optical tweezers setup, we found the fusion speeds of four types of droplets to differ by two orders of magnitude. The order of fusion speed correlates with the fluorescence of thioflavin T, which in turn reflects the macromolecular packing density inside droplets. Unstructured protein or polymer chains pack loosely and readily rearrange, leading to fast fusion. In contrast, structured protein domains pack more closely and have to break extensive contacts before rearrangement, corresponding to slower fusion. This molecular interpretation for disparate fusion speeds provides mechanistic insight into the assembly and aging of biomolecular droplets. 相似文献

建立了在线固相萃取-液相色谱-串联质谱(online SPE-LC-MS/MS)测定蘑菇中毒患者尿液中痕量α -鹅膏毒肽的分析方法。样品经甲酸酸化的乙腈-甲醇(5:1,v/v)沉淀蛋白质,反相液液微萃取去除样品提取液中的有机溶剂,毒素经ODS微柱(5 mm×2.1 mm,5 μm)在线SPE净化,XBridgeTM BEH C18 色谱柱(150 mm×3.0 mm,2.5 μm)分离,MS/MS测定。采用基于定量环的快速阀切换技术作为在线SPE和LC-MS/MS模块的接口,使得两个分离模块互相独立,无论是流动相还是压力,都不会互相干扰,保证了系统的稳定性;在线系统的精准净化,有效消除了后续质谱检测的基质效应,确保了尿液中痕量水平α -鹅膏毒肽的定性定量检测。尿液中α -鹅膏毒肽在0.1~50 μg/L范围内线性关系良好,相关系数(r 2 )为0.9983;检出限(LOD)为0.03 μg/L;α -鹅膏毒肽的加标(0.1、2.0和20 μg/L)平均回收率为84.3%~91.7%,相对标准偏差(RSD)为3.8%~7.2%。体内鹅膏毒肽代谢迅速,生物基质中痕量水平毒素的检测是其中毒实验室鉴定的主要难题,通过实际样品检测,证明该法操作简单,准确、灵敏;溶剂沉淀蛋白质和反相液液微萃取去除有机相和脂溶性基质的简单操作,可以作为水溶性毒素在线SPE-LC-MS/MS检测时快速且有效的配套前处理方法;基于在线SPE精准净化技术,可以实现尿液中α -鹅膏毒肽的高灵敏度测定(LOD为0.03 μg/L),解决了中毒时患者体内痕量水平α -鹅膏毒肽定性确证的难题,部分患者α -鹅膏毒肽中毒实验室鉴定的时间可以扩展到90 h以上;同时,痕量水平的定量检测技术,可以为中毒后迅速代谢的α -鹅膏毒肽在体内的剂量反应关系研究提供可靠的技术支撑。 相似文献

Efficient local gene transfection on a tissue scaffold is dependent on good cell-adhesion characteristics. In this work, the thermo-responsive gelatin-functionalized polycaprolactone (PCL) films were proposed for improvement of cell adhesion and intelligent recovery of gene-transfected cells. Functional copolymer brushes (PCL-g-P(NIPAAm-co-MAAS)) were first prepared via surface-initiated ATRP of N-isopropylacrylamide (NIPAAm) and methacrylic acid sodium salt (MAAS) from the initiator-funcationalized PCL surfaces. The pendant carboxyl end-groups of the PCL-g-P(NIPAAm-co-MAAS) surface were subsequently coupled with gelatin via carbodiimide chemistry to produce the thermo-responsive gelatin-functionalized PCL surface. The thermo-responsive gelatin-functionalized PCL film surface can improve cell adhesion and proliferation above the LCST of P(NIPAAm) without destroying cell detachment properties at lower temperatures. The dense transfected cells can be recovered simply by lowering culture temperature. The thermo-responsive gelatin-functionalized PCL films are potentially useful as intelligent adhesion modifiers for directing cellular functions within tissue scaffolds. 相似文献

Protein labeling with synthetic moieties remains in many cases a technically challenging or unresolved task. Two new and simple concepts are presented. In both approaches, a very short tag of only a few amino acids is prepared with the desired chemical modification and, in a second step, it is transferred to the protein of interest by protein trans‐splicing. For the amine‐tag, a recombinant intein fragment free of lysine residues was generated such that the amine group of the N terminus could be selectively modified with regular amine‐reactive reagents. Thus, standard bioconjugation procedures without any chemical synthesis could be applied without modification of lysines in the protein of interest. For the click‐tag, protein trans‐splicing was combined with unnatural amino acid mutagenesis and subsequent bioorthogonal side chain modification, as demonstrated for click chemistry using p‐azidophenylalanine. By the two‐step strategy, exposure of the protein of interest to the copper catalyst was avoided. 相似文献

Large (200 nm) poly(ethylene oxide)‐b‐poly(butadiene) polymer vesicles fuse into giant (>1 μm) vesicles with mild agitation in dilute aqueous NaCl solutions. This unusual effect is attributed to the salt‐induced contraction of the poly(ethylene oxide) corona, reducing steric resistance between vesicles and, with agitation, increasing the probability of contact between the hydrophobic cores of adjacent membranes. In addition, NaCl and agitation facilitated the creation of giant hybrid vesicles from much smaller homogeneous polymersomes and liposomes. Whereas lipid vesicles do not readily fuse with each other under the same circumstances, they did fuse with polymersomes to produce hybrid polymer/lipid vesicles. 相似文献

The 6,6‐quinolone scaffold of the viridicatin‐type of fungal alkaloids are found in various quinolone alkaloids which often exhibit useful biological activities. Thus, it is of interest to identify viridicatin‐forming enzymes and understand how such alkaloids are biosynthesized. Here an Aspergillal gene cluster responsible for the biosynthesis of 4′‐methoxyviridicatin was identified. Detailed in vitro studies led to the discovery of the dioxygenase AsqJ which performs two distinct oxidations: first desaturation to form a double bond and then monooxygenation of the double bond to install an epoxide. Interestingly, the epoxidation promotes non‐enzymatic rearrangement of the 6,7‐bicyclic core of 4′‐methoxycyclopenin into the 6,6‐quinolone viridicatin scaffold to yield 4′‐methoxyviridicatin. The finding provides new insight into the biosynthesis of the viridicatin scaffold and suggests dioxygenase as a potential tool for 6,6‐quinolone synthesis by epoxidation of benzodiazepinediones. 相似文献

This study focuses on the MCM-41 material (Mobil Composition of Matter). The MCM-41 nanomaterial presents higher physical properties such as pore sizes, surface areas and pore volumes. This material is usually synthesized by using laboratory reagents as silicate sources and aluminium source. These laboratory reagents are still expensive and toxic for large scale production. The main aim of this work is to resolve this problem and to replace these expensive laboratory reagents by more cost effective ones. The volclay and Algerian bentonite low-cost mass clay materials are used as silicate and acuminate sources separately by adopting an alkaline fusion process to extract both silicon and aluminium (1 kg of silicium and aluminium from volclay and Algerian bentonite cost around 0.03 and 0.01 € whereas the same amount of silicon from ludox and aluminium from sodium aluminates cost around 350 €). The synthesis of MCM-41 from bentonite was carried out by the hydrothermal method using the supernatants of bentonite (in the form of sodium silicate and sodium aluminate). On the basis of the data obtained from powder X-ray diffraction (XRD), transmission electron microscopy (TEM) and N2 adsorption and desorption, the results revealed that the properties of MCM-41 synthesized from Algerian bentonite and volclay separately depend on both elemental composition and mineral phase contents of the used bentonite. Pure and highly ordered hexagonal mesoporous MCM-41 with uniform pore sizes and a high specific surface area have been successfully synthesized without any phases which exist in natural bentonite.
